Melbourne Pathology operates 300+ collection centres across greater Melbourne and regional VIC for routine pathology, but semen analysis drop-off is restricted to the two sites above. Fertility-clinic on-site collection rooms exist at Melbourne IVF, Monash IVF, Genea, City Fertility, Adora Fertility, and other Melbourne fertility services — all need a fertility-specialist referral and are typically used as part of an IVF/ICSI workup rather than for stand-alone GP-referred testing. The Hospital fertility unit (public) and Royal Melbourne Hospital andrology service handle complex andrology referrals — typically by referral from a treating specialist rather than a primary-care GP.

Important WA-specific change. Effective 14 April 2024, PathWest (the WA public pathology service) no longer offers fertility analysis on semen samples, including samples for vasectomy reversal. PathWest continues post-vasectomy semen analysis only (metro patients deliver to PathWest, QEII Medical Centre, J Block Mon–Fri 8am–3pm; regional Mon–Thu only). For fertility testing, samples must go to one of the private providers below.

Public sector pathway — King Edward Memorial Hospital runs a Reproductive Medicine Service / Fertility Clinic for patients already in tertiary public-hospital care; pathway-specific instructions come from the treating clinician. PathWest no longer accepts fertility samples (cessation note above).

SA-specific note. Unlike Western Australia (where PathWest stopped fertility semen analysis in April 2024), SA Pathology — the South Australian public pathology service — still accepts fertility and post-vasectomy semen samples at four metropolitan laboratories. The public service (Territory Pathology, based at Royal Darwin Hospital) primarily supports public hospital diagnostics rather than community fertility/post-vasectomy testing. Everything you need to know about semen analysis

Semen analysis is time-sensitive. Once collected, the sample must reach the laboratory and be examined within 40 minutes. Sperm motility and survival decline rapidly outside the body — so the lab you choose, the way you transport the sample, and your timing all directly affect whether the result is valid. Every lab needs a phone booking before you collect; no lab accepts walk-in semen samples without an appointment.

You'll need a referral first. Clinic365 currently issues semen-analysis pathology referrals through a Specialist GP telehealth consultation only — the GP calls you, takes a brief history, and sends the pathology referral by SMS. There is no walk-in or self-directed online intake for sperm tests at the moment.

Two different reasons people are tested. Fertility testing assesses sperm count, motility, morphology, and volume to investigate why a couple isn't conceiving. Post-vasectomy testing checks whether sperm have cleared from the ejaculate after a vasectomy, before you can stop using contraception. The collection process is similar, but timing and abstinence rules differ.

For fertility testing

Abstain for 2–7 days. Don't ejaculate (no intercourse and no masturbation) for at least two full days before your test, and no longer than seven days. Three to five days is the sweet spot. Record the exact number of days on the container — the lab uses this to interpret your result.

Get a sterile container from the lab first. Don't use a jar from home — household containers contain residues that kill sperm. The pathology centre will give you the correct sterile container when you book your appointment.

The collection itself. Wash your hands and penis with soap, then rinse thoroughly so all soap is removed (soap kills sperm). Collect the sample by masturbation, directly into the container — no condoms, no lubricants, no saliva. Catch the entire ejaculate, including the very first part (the first portion contains most of the sperm). Screw the lid on tightly. Label the container with your full name, date of birth, and the exact date and time of collection.

Keep the sample warm. Carry the container in an inside shirt or trouser pocket, close to your body. Don't refrigerate it. Don't leave it in a cold or hot car. Body temperature is the goal.

Get to the lab fast. Within 40 minutes of collection. If you can't make that window from home, ask the lab whether they have a private collection room you can use on-site — most major fertility-aligned labs do.

Cost and. Fertility semen analysis is generally not — gap fees vary by provider. Confirm pricing when you book. Results are usually available to your referring doctor within 5 working days.

For post-vasectomy testing (PVSA)

Wait at least 12 weeks AND 20 ejaculations. Both conditions must be met before your first post-vasectomy test. Continue using contraception until your doctor sends written clearance — a vasectomy is not effective until confirmed clear by lab testing.

Abstinence is usually not needed. Confirm with your vasectomy surgeon — for most post-vasectomy tests, the 2–7 day abstinence rule does not apply.

Collection is otherwise the same as fertility testing. Sterile container from the lab, masturbation only, no condoms or lubricants, complete ejaculate, label clearly, keep warm, deliver promptly.

If sperm are still present. A small percentage of post-vasectomy tests show residual sperm at 12 weeks. The lab will report whether they're motile (moving) or non-motile (not moving) — your doctor will advise next steps. Most cases simply need another month and another 20 ejaculations before a repeat test.

Common reasons your test gets rejected or repeated

Sample dropped off after the time limit (kept too long or kept cold). Container wasn't sterile or wasn't supplied by the lab. Condom or lubricant used during collection. Part of the ejaculate was lost (often the first portion). Abstinence period was outside the 2–7 day window. Patient information form not completed. Phone booking wasn't made before drop-off.
